North Yorkshire Police said the rider of a Honda motorcycle, in his 40s, sustained serious leg injuries in the incident on Catterick Road, Colburn, near Catterick Garrison, shortly after 3.30pm.
He was treated by paramedics at the scene before being taken to James Cook University Hospital, in Middlesbrough, by air ambulance. The driver of the Ford Focus car was not believed to have been injured.
Motorists were initially urged to avoid the area due to heavy traffic congestion.

The road reopened around 6.45pm.
Witnesses are asked to call police on 101, and speak to the Force Control Room, quoting reference NYP-11092015-0325
