The store, which was previously occupied by BHS, opened in November 2017.
Prior to the company moving in, the unit had been empty since the closure of BHS in 2016.
A spokeswoman for the company said it had ceased trading on Saturday and was looking for a new location in the city centre.
She said: “The store ceased trading on Saturday but we have such a loyal customer base in Durham we are looking for a new location.”
She added the lease had expired on the current premises in Prince Bishops.
M&Co is looking for new city centre premises but has not yet signed a deal.
