The road was closed for several hours following the crash and queues built between Crindle Carr Lane and the B1448 Northallerton Road.
The road reopened and police left the scene at around 1pm.
At the time of the incident, North Yorkshire Police said: "The A168 at Thornton le Street is closed following a serious collision this morning. The road will be closed for some time while we work at the scene."
Police confirmed that two women - both the drivers - had been taken to hospital.
Meawhile, two drivers were reported for driving through the road closure signs while emergency services worked at the scene.
This led to an appeal from the police urging motorists to respect the closure signs and to not put the lives of emergency service personnel at risk.
